As for the enemies, you will need to take them down so choose a Weapon from the rack and go through the basics of the game in your one vs one duel.
After the battle the area will come under attack so get through taking out the Yokai to where you meet up with Hattori Hanzo. Grab a Ninjitsu weapon then clear out the Yokai, follow Hanzo up to the balcony using the ladder he drops for you then head down into the well.
Like I said earlier, the boss in this mission is one you’re supposed to die to so do what you can but ultimately you will more than likely lose which then triggers a cut-scene concluding the mission.
